My research has started from development of new synthetic methodology. The new methods have been successfully applied to e.g. total syntheses of several natural products (e.g. jaspine B, nojirimycin analogues, amaminol, sphingolipids), as well as short and efficient syntheses of active pharmaceutical agents (e.g. pregabalin). These syntheses necessitated in-depth investigations in the underlying mechanisms and theory of the reactions involved. This deeper understanding again allowed us to invent powerful green technologies such as: Cu-catalyzed air-oxidation of alcohols; Cu-mediated hydrosilylations of unsaturated carbonyl compounds, (organo)catalytic conjugate additions. and the first organocatalytic IMDA in 2005. The underlying theme in all these methodology/small molecule investigations has been a long term goal to achieve practical syntheses of complex organic molecules to interrogate the biological systems. I have chosen two major important targets: cellular control of the cell cycle through the action of the protein phosphorylation mechanisms (calyculins through protein phosphatases), and sequence selective recognition of DNA stretches with synthetic structurally defined man-designed probe molecules. BACKGROUND
I have been highly motivated from early stages of my academic development: I was among the youngest ever PhD's in Finland and at the age of 26 (including one year of industrial experience and one year of military service), I was the youngest chemistry doctoral candidate of Helsinki University of Technology. I then decided to look for the best possible further education I could obtain, and moved to the University of California, Berkeley, to do a two year post-doc with Professor Henry Rapoport, an eminent pioneer in asymmetric synthesis of natural products. In 1985 I returned to Finland for a two year project in the pharmaceutical industry (Orion Corporation – Fermion). My task was to initiate a new drug development group, an industrial R&D function then in its infancy in Finland, which led me to learn more of drug development and computer aided drug design: my group was among the pioneers to implement chemical database management systems (then MDL) and molecular modeling (Clark Still's MacroModel) into routine daily work of a synthetic medicinal chemist. We also actively worked to develop computerized methods, and one member of my group spent half a year in the research group of Professor Corwin Hansch, a pioneer in QSAR. The student eventually became the first one in Scandinavia to defend his thesis on 3D QSAR methods. During this time I was also trained as a group leader and my responsibility was to direct a group of 10-15 people of varied academic backgrounds. After the two years in industry, I decided to move back to academia, and upon the encouragement of Professor Rapoport, I returned to California in 1987. Looking for academic jobs both in the States and Europe at the end of the 80's was very frustrating, and after frozen offers from MIT (Department of Applied Biological Sciences, which was closed) and University of Arizona, Tucson, I landed on my first independent academic job as a lecturer at the University of Surrey in 1989. In 1991 I had the opportunity to return to Finland, as a full professor at the University of Oulu. After Oulu, at the turn of the millennium, I was a co-founder of a small company, and acted as the CEO and CSO for a combined period of nearly two years before taking up the full time responsibilities at my alma mater, Helsinki University of Technology, now Aalto University, in 2000 as the Professor of Organic Chemistry.
